Description
Organoids and cancer spheroids are often too opaque for optical imaging by single-sided illumination. Rotating the sample for multi-angle illumination solves this, enabling 3D tomographic reconstruction of the refractive index. Optical and acoustic forces can both manipulate microscopic samples in suspension without contact, but heavier samples require acoustic levitation. Tuning standing MHz ultrasound waves from various directions allows one to rotate a sample within a microfluidic chamber. The method is non-invasive and avoids the "missing-cone" artifact problem. Examples shown include 3D reconstructions of a levitated zebrafish larva via Optical Coherence Tomography (OCT) and of cell clusters via Optical Diffraction Tomography (ODT).
